P-52: Brain-Derived Neurotrophic Factor Promotes The Development of Human Ovarian Early Follicles during Growth In Vitro

چکیده مقاله:
Background Cryopreservation of ovarian cortex is increasingly used to preserve fertility before cancer therapy. Recently, studies show that Brain-derived neurotrophic factor (BDNF) may be involved in oocyte maturation. Brain-derived neurotrophic factor (BDNF) is member of neurotrophin family that has anti-apoptotic effects on nervous system. Recent researches show that it also plays key role in female reproductive system such as nuclear and cytoplasmic maturation and embryo development. Therefore, the present study was conducted to evaluate the effects of BDNF on morphology, viability and apoptosis of primordial follicles, when it was added to in vitro culture media. MaterialsAndMethods Cortical tissue strips from normal pregnant (n=8) women were vitrified. After two weeks, thawed cortical strip was divided to 3 groups as following: FSH, FSH + BDNF (100ng) and control (without FSH). Subsequently, cortical strip was cultured in a (α-MEM) medium for two weeks. Then ovarian fragments were analyzed for morphology (Hematoxylin and eosin, H & E), and incidence of apoptosis (TUNEL kit). Hormones (Estradiol, Anti mullerian hormone and progestron) concentration in culture medium and viability were evaluated using ELISA kit and Calcein-AM Ethidium homodimer-1 respectively. Results Our data indicates that the ratio of primary and secondary follicles to primordial significantly increased in BDNF+ FSH group compared with the other groups. Estradiol and progestron concentration were also significantly increased in BDNF+ FSH group compared with the other groups. Moreover, the percentage of apoptosis in BDNF+ FSH group was decreased significantly in comparison to the other groups. Viability was also significantly higher in BDNF+ FSH group compared to the other groups. Conclusion This data suggests that combination of BDNF and FSH have beneficial effects on human ovarian early follicles growth in vitro.

STUDY QUESTION What is the effect of basic fibroblast growth factor (bFGF) on the growth of individual early human follicles in a three-dimensional (3D) culture system in vitro? SUMMARY ANSWER The addition of 200 ng bFGF/ml improves human early follicle growth, survival and viability during growth in vitro. متن کاملOvarian brain-derived neurotrophic factor (BDNF) promotes the development of oocytes into preimplantation embryos.
Optimal development of fertilized eggs into preimplantation embryos is essential for reproduction. Although mammalian oocytes ovulated after luteinizing hormone (LH) stimulation can be fertilized and promoted into early embryos in vitro, little is known about ovarian factors important for the conditioning of eggs for early embryo development. Because LH interacts only with ovarian somatic cells...

Background Fertility preservation is an important part of scientific study in the field of reproductive medicine. Ovarian cryopreservation and in vitro follicles culture provide option for fertility conservation in older women or cancer patients. Basic fibroblast growth factor (bFGF) or FGF-2, is member of fibroblast growth factors family which play critical roles in cell migration proliferatio...
متن کاملP-219: GDF9, BMP15 and Their Receptors Expression During In Vitro Culture of Mouse Vitrified Ovarian Tissue Derived Preantral Follicles
Background: In vitro culture (IVC) of isolated preantral follicles from cryopreserved ovarian tissue might be an efficient method for enhancing mature oocytes in patients who are exposed to infertility. Materials and Methods: Ovaries of 13-day old NMRI mice were removed and randomly placed into control, needle immersed (NIV) and solid surface vitrification (SSV) groups. For vitrification, ovarie...
متن کاملGlial-derived neurotrophic factor promotes ovarian primordial follicle development and cell-cell interactions during folliculogenesis.
Female fertility is determined in part by the size and development of the primordial follicle pool. The current study investigates the role of glial cell-line-derived neurotrophic factor (GDNF) in the regulation of primordial follicle development in the ovary. Ovaries from 4-day-old female rat pups were maintained in organ culture for 10 days in the absence (control) or presence of GDNF or kit ...
متن کاملP-70: The Effect of Hydrostatic Pressure on Parthenogenetic Activation of Mouse Oocytes Derived from In vitro Grown Ovarian Follicles
Background: Parthenogenesis is the production of an embryo from a female gamete in the absence of any contribution from a male gamete, with or without the eventual development into an adult. In vivo, mammalian parthenogenesis is a rare event. Parthenogenesis can be efficiently induced in vitro with a variety of mechanical, chemical, and electrical stimuli in several species. Hydrostatic pressur...
